Abstract:
A simple, economical and efficient purification method for citrus yellow vein clearing virus (CYVCV) particles was developed. Abundant viral particles could be obtained with one sucrose cushion ultracentrifugation step of 2.5 h. Characterization of purified virus assessed by electron microscopy, SDS-PAGE and western blot revealed the typical flexuous particles of CYVCV without any visible contamination or degradation. Furthermore, the purified virus particles were biologically active and initiated successful infection upon inoculation to different citrus cultivars.
